Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.1354T>A (p.Cys452Ser), citing Ambry Variant Classification Scheme 2023: The p.C452S variant (also known as c.1354T>A), located in coding exon 5 of the AXIN2 gene, results from a T to A substitution at nucleotide position 1354. The cysteine at codon 452 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
