NM_000384.3(APOB):c.1219C>A (p.Leu407Met) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L407M variant (also known as c.1219C>A), located in coding exon 10 of the APOB gene, results from a C to A substitution at nucleotide position 1219. The leucine at codon 407 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
