NM_004336.5(BUB1):c.3215G>A (p.Arg1072Lys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1072K variant (also known as c.3215G>A), located in coding exon 25 of the BUB1 gene, results from a G to A substitution at nucleotide position 3215. The arginine at codon 1072 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
