Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.23C>T (p.Thr8Ile), citing Ambry Variant Classification Scheme 2023: The p.T8I variant (also known as c.23C>T), located in coding exon 2 of the RECQL gene, results from a C to T substitution at nucleotide position 23. The threonine at codon 8 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
