Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000722.4(CACNA2D1):c.35C>T (p.Thr12Ile), citing Ambry Variant Classification Scheme 2023: The p.T12I variant (also known as c.35C>T), located in coding exon 1 of the CACNA2D1 gene, results from a C to T substitution at nucleotide position 35. The threonine at codon 12 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
