NM_000722.4(CACNA2D1):c.118A>G (p.Met40Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M40V variant (also known as c.118A>G), located in coding exon 2 of the CACNA2D1 gene, results from an A to G substitution at nucleotide position 118. The methionine at codon 40 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
