Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001370259.2(MEN1):c.25A>C (p.Thr9Pro), citing Ambry Variant Classification Scheme 2023: The p.T9P variant (also known as c.25A>C), located in coding exon 1 of the MEN1 gene, results from an A to C substitution at nucleotide position 25. The threonine at codon 9 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
