NM_002880.4(RAF1):c.68A>G (p.Asp23Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D23G variant (also known as c.68A>G), located in coding exon 1 of the RAF1 gene, results from an A to G substitution at nucleotide position 68. The aspartic acid at codon 23 is replaced by glycine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

Protein context (NP_002871.1, residues 13-33): NGFGFKDAVF[Asp23Gly]GSSCISPTIV
