Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.8317C>T (p.Arg2773Cys), citing Ambry Variant Classification Scheme 2023: The p.R2344C variant (also known as c.7030C>T), located in coding exon 26 of the OBSCN gene, results from a C to T substitution at nucleotide position 7030. The arginine at codon 2344 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
