NM_001386125.1(OBSCN):c.12020A>G (p.Asp4007Gly) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D3578G variant (also known as c.10733A>G), located in coding exon 40 of the OBSCN gene, results from an A to G substitution at nucleotide position 10733. The aspartic acid at codon 3578 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
