NM_000548.5(TSC2):c.3910A>C (p.Ser1304Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1304R variant (also known as c.3910A>C), located in coding exon 32 of the TSC2 gene, results from an A to C substitution at nucleotide position 3910. The serine at codon 1304 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
