NM_000264.5(PTCH1):c.4310G>T (p.Cys1437Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C1437F variant (also known as c.4310G>T), located in coding exon 23 of the PTCH1 gene, results from a G to T substitution at nucleotide position 4310. The cysteine at codon 1437 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
