NM_001378454.1(ALMS1):c.9910T>A (p.Ser3304Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S3305T variant (also known as c.9913T>A), located in coding exon 13 of the ALMS1 gene, results from a T to A substitution at nucleotide position 9913. The serine at codon 3305 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
