Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.8799C>G (p.Cys2933Trp), citing Ambry Variant Classification Scheme 2023: The p.C2934W variant (also known as c.8802C>G), located in coding exon 10 of the ALMS1 gene, results from a C to G substitution at nucleotide position 8802. The cysteine at codon 2934 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
