Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001367624.2(ZNF469):c.586T>G (p.Tyr196Asp), citing Ambry Variant Classification Scheme 2023: The p.Y196D variant (also known as c.586T>G), located in coding exon 1 of the ZNF469 gene, results from a T to G substitution at nucleotide position 586. The tyrosine at codon 196 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
