NM_006361.6(HOXB13):c.268T>G (p.Cys90Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C90G variant (also known as c.268T>G), located in coding exon 1 of the HOXB13 gene, results from a T to G substitution at nucleotide position 268. The cysteine at codon 90 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
