Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.812A>G (p.Glu271Gly), citing Ambry Variant Classification Scheme 2023: The p.E271G variant (also known as c.812A>G), located in coding exon 2 of the HOXB13 gene, results from an A to G substitution at nucleotide position 812. The glutamic acid at codon 271 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
