Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4668C>G (p.Ser1556Arg), citing Ambry Variant Classification Scheme 2023: The p.S1510R variant (also known as c.4530C>G), located in coding exon 40 of the KIF1B gene, results from a C to G substitution at nucleotide position 4530. The serine at codon 1510 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
