Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.5146A>C (p.Ser1716Arg), citing Ambry Variant Classification Scheme 2023: The p.S1670R variant (also known as c.5008A>C), located in coding exon 44 of the KIF1B gene, results from an A to C substitution at nucleotide position 5008. The serine at codon 1670 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
