Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4913T>C (p.Leu1638Pro), citing Ambry Variant Classification Scheme 2023: The p.L1592P variant (also known as c.4775T>C), located in coding exon 42 of the KIF1B gene, results from a T to C substitution at nucleotide position 4775. The leucine at codon 1592 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
