NM_017636.4(TRPM4):c.2502C>A (p.Ser834Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S834R variant (also known as c.2502C>A), located in coding exon 17 of the TRPM4 gene, results from a C to A substitution at nucleotide position 2502. The serine at codon 834 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been reported in a sudden unexplained death cohort; however, clinical details were limited (Subbotina E et al. Forensic Sci Int, 2018 Dec;293:37-46). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
