This variant lies in the DNAAF5 gene (transcript NM_017802.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1931, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.1931+2T>C intronic variant results from a T to C substitution two nucleotide after coding exon 9 of the DNAAF5 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and will result in the creation or strengthening of a novel splice donor site.
