NM_001903.5(CTNNA1):c.1304A>T (p.Asn435Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N435I variant (also known as c.1304A>T), located in coding exon 9 of the CTNNA1 gene, results from an A to T substitution at nucleotide position 1304. The asparagine at codon 435 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
