Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.1347T>G (p.Asn449Lys), citing Ambry Variant Classification Scheme 2023: The p.N449K variant (also known as c.1347T>G), located in coding exon 10 of the FBN2 gene, results from a T to G substitution at nucleotide position 1347. The asparagine at codon 449 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
