Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_006073.4(TRDN):c.1135+1G>A, citing Ambry Variant Classification Scheme 2023: The c.1135+1G>A intronic variant results from a G to A substitution one nucleotide after coding exon 14 of the TRDN gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site and may result in the creation or strengthening of a novel splice acceptor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, this alteration does not impact the predominant cardiac isoform of TRDN (NM_001256021.1; Kobayashi YM et al. J. Biol. Chem., 1999 Oct;274:28660-8). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
