Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.1998-81A>C, citing Ambry Variant Classification Scheme 2023. This variant lies in the TSC1 gene (transcript NM_000368.5) at 81 bases into the intron immediately before coding-DNA position 1998, where A is replaced by C. Submitter rationale: The c.1998-81A>C intronic variant results from an A to C substitution 81 nucleotides upstream from coding exon 14 in the TSC1 gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. RNA studies have demonstrated that this alteration results in a splice defect; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
