Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_020975.6(RET):c.1612A>G (p.Thr538Ala), citing Ambry Variant Classification Scheme 2023: The p.T538A variant (also known as c.1612A>G), located in coding exon 8 of the RET gene, results from an A to G substitution at nucleotide position 1612. The threonine at codon 538 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
