NM_000492.4(CFTR):c.682G>A (p.Gly228Ser) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G228S variant (also known as c.682G>A), located in coding exon 6 of the CFTR gene, results from a G to A substitution at nucleotide position 682. The glycine at codon 228 is replaced by serine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
