Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001365276.2(TNXB):c.1170C>G (p.Cys390Trp), citing Ambry Variant Classification Scheme 2023: The p.C390W variant (also known as c.1170C>G), located in coding exon 2 of the TNXB gene, results from a C to G substitution at nucleotide position 1170. The cysteine at codon 390 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
