Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001231.5(CASQ1):c.376G>A (p.Val126Met), citing Ambry Variant Classification Scheme 2023: The p.V126M variant (also known as c.376G>A), located in coding exon 3 of the CASQ1 gene, results from a G to A substitution at nucleotide position 376. The valine at codon 126 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
