NM_001148.6(ANK2):c.11086G>C (p.Glu3696Gln) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E3696Q variant (also known as c.11086G>C), located in coding exon 42 of the ANK2 gene, results from a G to C substitution at nucleotide position 11086. The glutamic acid at codon 3696 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
