NM_001378454.1(ALMS1):c.3635C>G (p.Pro1212Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P1213R variant (also known as c.3638C>G), located in coding exon 8 of the ALMS1 gene, results from a C to G substitution at nucleotide position 3638. The proline at codon 1213 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
