Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.5095A>C (p.Ser1699Arg), citing Ambry Variant Classification Scheme 2023: The p.S1653R variant (also known as c.4957A>C), located in coding exon 43 of the KIF1B gene, results from an A to C substitution at nucleotide position 4957. The serine at codon 1653 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
