Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002878.4(RAD51D):c.214T>C (p.Tyr72His), citing Ambry Variant Classification Scheme 2023: The p.Y72H variant (also known as c.214T>C), located in coding exon 3 of the RAD51D gene, results from a T to C substitution at nucleotide position 214. The tyrosine at codon 72 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
