NM_003579.4(RAD54L):c.1963C>T (p.His655Tyr) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H655Y variant (also known as c.1963C>T), located in coding exon 17 of the RAD54L gene, results from a C to T substitution at nucleotide position 1963. The histidine at codon 655 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
