Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_144997.7(FLCN):c.557G>T (p.Trp186Leu), citing Ambry Variant Classification Scheme 2023: The p.W186L variant (also known as c.557G>T), located in coding exon 3 of the FLCN gene, results from a G to T substitution at nucleotide position 557. The tryptophan at codon 186 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
