NM_006904.7(PRKDC):c.2981G>T (p.Trp994Leu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W994L variant (also known as c.2981G>T), located in coding exon 26 of the PRKDC gene, results from a G to T substitution at nucleotide position 2981. The tryptophan at codon 994 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
