Uncertain significance — the classification assigned by Ambry Genetics to NM_006393.3(NEBL):c.1447G>A (p.Glu483Lys), citing Ambry Variant Classification Scheme 2023: The p.E483K variant (also known as c.1447G>A), located in coding exon 14 of the NEBL gene, results from a G to A substitution at nucleotide position 1447. The glutamic acid at codon 483 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
