Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.3318A>C (p.Glu1106Asp), citing Ambry Variant Classification Scheme 2023: The p.E1089D variant (also known as c.3267A>C), located in coding exon 18 of the PALLD gene, results from an A to C substitution at nucleotide position 3267. The glutamic acid at codon 1089 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
