Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003001.5(SDHC):c.365T>G (p.Phe122Cys), citing Ambry Variant Classification Scheme 2023: The p.F122C variant (also known as c.365T>G), located in coding exon 5 of the SDHC gene, results from a T to G substitution at nucleotide position 365. The phenylalanine at codon 122 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
