Uncertain significance — the classification assigned by Ambry Genetics to NM_012282.4(KCNE5):c.20A>G (p.Gln7Arg), citing Ambry Variant Classification Scheme 2023: The p.Q7R variant (also known as c.20A>G), located in coding exon 1 of the KCNE5 gene, results from an A to G substitution at nucleotide position 20. The glutamine at codon 7 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
