Pathogenic for DICER1-related tumor predisposition — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_177438.3(DICER1):c.4605_4606del (p.Cys1535fs), citing Invitae Variant Classification Sherloc (09022015): Loss-of-function variants in DICER1 are known to be pathogenic (PMID: 19556464, 21266384). This variant has been observed in an individual affected with pleuropulmonary blastoma (PMID: 26925222) and has also been observed to be de novo in an individual affected with DICER1 syndrome (PMID: 28960912). ClinVar contains an entry for this variant (Variation ID: 254337). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Cys1535Trpfs*3) in the DICER1 gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic.
