Likely pathogenic for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000535.6(PMS2):c.1145-?_2174+?dup1030, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross duplication of the genomic region encompassing exons 11 and 12 of the PMS2 gene. While the exact position of the duplicated exons cannot be determined from this data, the duplicated copy of this region is likely in tandem and may result in an absent or disrupted protein product. Truncating variants, including gross duplications, in PMS2 are known to be pathogenic (PMID: 21376568, 24362816). A similar duplication of exons 11 and 12 has been reported in the literature in patients subjected to hereditary cancer testing (PMID: 25512458).
