NM_000251.2(MSH2):c.-125_645+?del was classified as Pathogenic for Hereditary nonpolyposis colorectal neoplasms by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 1-3 of the MSH2 gene, which includes the initiator codon. This deletion includes the translation initiation methionine and extends beyond both edges of the assayed region, and therefore the 5' and 3' boundaries of this event are not known, although the 3' end is somewhere in intron 3. It is expected to result in a disrupted or absent protein product. Deletions of exons 1-3 have been reported in several affected patients and families, and are clearly defined as Lynch syndrome causative alleles (PMID: 19930554, 12373605, 19250818). For these reasons, this variant has been classified as Pathogenic.
