Likely pathogenic for Familial cancer of breast — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024675.3(PALB2):c.1685-?_2748+?dup, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross duplication of the genomic region encompassing exons 5-7 of the PALB2 gene. While the exact position of the duplicated exons cannot be determined from this data, the duplicated copy of this region is likely in tandem and may result in an absent or disrupted protein product. While gross duplications in PALB2 are known to be pathogenic (PMID: 24136930), this particular duplication has not been reported previously in the literature. For these reasons, this variant has been classified as Likely Pathogenic.
