Pathogenic for Tuberous sclerosis 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000548.3(TSC2):c.1600-?_1716+?del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 16 of the TSC2 gene. This deletion extends beyond the edge of the assayed exon, and the exact 5' and 3' boundaries of this event are not known but most likely lie within introns 15 and 16. This leads to an in-frame deletion, preserving the integrity of the reading frame. Loss-of-function variants including gross deletions in TSC2 are known to be pathogenic. Deletions encompassing exon 16 have been reported in the literature in individuals affected with tuberous sclerosis complex (PMID: 16114042, 25782670) and in affected individuals in the Leiden Open-source Variation Database (PMID: 21520333). Deletions of exon 16 are known as deletions of exon 15 in the literature. For these reasons, this variant has been classified as Pathogenic.
