NM_000051.3(ATM):c.8269-?_8671+?del was classified as Pathogenic for Ataxia-telangiectasia syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 57-59 of the ATM gene. This deletion removes both exons 57 and 58, as well as the first 44 nucleotides of exon 59. Although the impact of this deletion on splicing is uncertain, it likely creates a premature translational stop signal resulting in an absent or disrupted protein product. While this particular variant has not been reported in the literature, gross deletions and truncating variants in ATM are known to be pathogenic (PMID: 25614872, 23807571). For these reasons, this variant has been classified as Pathogenic.
