Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.20623C>G (p.His6875Asp), citing Ambry Variant Classification Scheme 2023: The p.H5918D variant (also known as c.17752C>G), located in coding exon 73 of the OBSCN gene, results from a C to G substitution at nucleotide position 17752. The histidine at codon 5918 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
