Pathogenic for Early-infantile DEE — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001330260.2(SCN8A):c.779T>C (p.Phe260Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with serine at codon 260 of the SCN8A protein (p.Phe260Ser). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and serine. For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has been observed in individual(s) with clinical features of early infantile epileptic encephalopathy (PMID: 25568300, Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 253279). This variant is not present in population databases (ExAC no frequency).
