Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.3749G>A (p.Arg1250His), citing Ambry Variant Classification Scheme 2023: The p.R1250H variant (also known as c.3749G>A), located in coding exon 19 of the MYLK gene, results from a G to A substitution at nucleotide position 3749. The arginine at codon 1250 is replaced by histidine, an amino acid with highly similar properties. This alteration has been reported in a thoracic aortic aneurysm and dissection (TAAD) cohort (Weerakkody R et al. Genet Med, 2018 Nov;20:1414-1422). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
